Get title in front-matter

For a manuscript of ours I changed the front matter to send an 'adapted' email to the corresponding author, and I'm using the {{manubot.ci_source.repo_slug}} for setting the subject.

The comment at the beginning of the file mentions that "Pandoc automatically inserts title from metadata.yaml, so it is not included in this template", but would it nonetheless be possible to somehow get the title into the template to generate a "nice" subject on that email?

That information should be available for you to use in the template. The output branch shows the variables that are collected, e.g. https://github.com/manubot/rootstock/blob/output/variables.json

The existing template uses the manubot section of the JSON. There is also a pandoc section for the variables pandoc sets. Try pandoc.title.
